Objectives:

i) To develop clinical techniques for application of Geristore to Peri-implantitis patients; ii) Cell culture studies and characterisation of Geristore .

Methods:

Two patients with four implants affected by peri-implantitis were treated. Probing depths were 6-8mms, with bleeding on probing and exudation. At surgery, after debridement, implant surfaces were disinfected (0.2% chlorhexidine). Bone defects with intra bony component were filled with deprotinised bovine bone (Geistlich, BioOss) and covered with porcine collagen membrane (Geistlich, BioGide). The supra-bony parts of the exposed implant surface were treated with Geristore and flaps replaced and sutured.

ii) A Geristore specimen was characterised using SEM and EDX. Flouride/ ion release studies were carried out over 1 month using ICPMS and a fluoride electrode. Undifferentiated human mesenchymal stem cells were grown in culture (37 oC) in a humid, 5% CO2 atmosphere, in Dulbecco's Modified Essential Medium (DMEM) supplemented with 10% foetal bovine serum 2 mmol/L L-glutamine, 100 microgramme/mL penicillin, and 100 microgramme/mL streptomycin and seeded onto Geristore samples at a density of 5000 cells/sample. 10 Geristore samples and 10 controls were prepared and incubated (3 days). Alkaline phosphatase activity and DNA content assays were used to assess cell differentiation and proliferation.
